| 生信分析胃癌中MAPK相关基因表达及IL1A的免疫意义 |

| 为在MAPK相关基因中寻找胃癌有关生物标志物,通过TCGA,MSigDB数据库、R包limma和wilcoxon检验对胃癌中MAPK相关基因进行差异表达分析;通过R包survival进行生存分析;通过STRING数据库构建PPI网络;通过Metascape进行富集分析;通过cibersort计算胃癌免疫细胞浸润水平。最后,使用实时荧光定量PCR(RT-qPCR)在20例胃癌及癌旁正常组织样本中验证差异基因的表达。结果发现胃癌中MAPK相关基因中上调基因27个,下调基因59个;生存分析发现MAPK相关基因IL1A、MAP4K4、PDGFRB基因在胃癌组织中高表达并且影响胃癌预后。PPI网络显示差异基因表达蛋白具有广泛的相互作用;GO和KEGG功能分析显示IL1A、MAP4K4、PDGFRB基因主要与EGFR tyrosine kinase inhibitor resistance,MAPK signaling pathway等通路密切相关。胃癌免疫细胞浸润分析发现IL1A与肥大细胞浸润呈显著相关。RT-qPCR验证中,IL1A、MAP4K4、PDGFRB在胃癌组织中表达(1.90±0.34)、(2.10±0.44)、(2.08±0.40)均高于正常组织(0.90±0.16)、(0.96±0.18)、(1.08±0.20);由此得出结论:胃癌中MAPK相关基因IL1A、MAP4K4、PDGFRB存在差异表达且与预后相关,其中IL1A可能作为免疫治疗的潜在生物标志物。 |

| To search for biomarkers related to gastric cancer in MAPK related genes. Differential expression of MAPK-related genes in gastric cancer is analyzed by TCGA, MSigDB database, R-packet limma and wilcoxon test. Survival analysis is performed through R packet survival. Construct PPI network through STRING database. Enrichment analysis is performed by Metascape. The infiltration level of gastric cancer immune cells is calculated by cibersort. Finally, real-time fluorescence quantitative PCR (RT-qPCR) is used to verify the expression of differential genes in 20 gastric cancer and adjacent normal tissue samples. Among the MAPK related genes in gastric cancer, 27 are upregulated and 59 are downregulated. Survival analysis revealed that MAPK related genes IL1A, MAP4K4, and PDGFRBare highly expressed in gastric cancer tissue and affect the prognosis of gastric cancer. The PPI network showed that differentially expressed gene proteins have extensive interactions. GO and KEGG functional analysis show that the IL1A, MAP4K4, and PDGFRB genes are closely related to EGFR tyrosine kinase inhibitor resistance, MAPK signaling pathway, and other pathways. Analysis of immune cell infiltration in gastric cancer revealed a significant correlation between IL1Aand mast cell infiltration. In RT qPCR validation, the expression of IL1A, MAP4K4, and PDGFRBin gastric cancer tissue (1.90±0.34), (2.10±0.44) , (2.08±0.40) was higher than in normal tissue (0.90±0.16), (0.96±0.18), (1.08±0.20). MAPK related genes IL1A, MAP4K4, and PDGFRBare differentially expressed in gastric cancer and are associated with prognosis. Among them, IL1A may serve as a potential biomarker for immunotherapy. |
