普氏野马GH基因的克隆及生物信息学分析

摘要:
普氏野马(Equus przewalskii)属国家 I 级保护动物,是现存的马中唯一的的野生亚种。生长激素(Growth Hormone,GH)是大脑垂体前叶分泌的一种多肽类激素,对动物个体的生长、发育的影响极其明显。为了更好的保护普氏野马,提高其野外生存与繁殖能力,本研究利用PCR技术首次获得了普氏野马 GH 基因,并利用生物信息学方法对其编码的蛋白质结构和功能进行分析和预测,旨在探明普氏野马生长激素(Growth Hormone, GH)基因的结构特点以及其编码蛋白质的结构与功能。结果显示,普氏野马 GH 基因所在基因组DNA全长为1 577 bp,经BLAST比对后,发现普氏野马与家马(GenBank: EU939446.1)GH 基因序列相似性高达99.68%,属于同源序列。该基因编码蛋白质的氨基酸数量为152个,分子量为17 638.54 Da,理论等电点为9.01,带负电荷的残基总数(Asp+Glu)为19个,正电荷残基总数(Arg+Lys)为23个,分子式为C790H1274N216O226S7。普氏野马GH蛋白属于细胞外蛋白,含有生长激素样结构域,不具有相关跨膜结构,其含有信号肽的可能性为0.35%。这为普氏野马GH基因的开发利用奠定了分子基础。

Cloning and bioinformatics analysis of GH gene from Equus przewalskii

Abstract:
Equus przewalskiibelongs to the national first-class protected wild animals. It is the only surviving wild subspecies of horse. Growth hormone (GH) is a polypeptide hormone secreted by the anterior pituitary gland, which has an obvious effect on the growth and development of animals. In order to better protect the wild horse and improve its wild survival and breed ability, the GH gene of Equus przewalskiiwas obtained by PCR for the first time, and the structure and function of its encoded protein were analyzed and predicted by bioinformatics method, so as to explore the structural characteristics of GH gene and the structure and function of its encoded protein. Results showed that the total length of genomic DNA of GH gene of Equus przewalskiiwas 1577 bp. After BLAST comparison, it was found that the sequence similarity of GH gene between Equus Przewalskiiand Equus caballus(GenBank: EU939446.1) was as high as 99.68%, indicating that they belong to homologous sequence. The number of amino acids of the protein encoded by the gene was 152, the molecular weight was 17 638.54 Da, the theoretical isoelectric point was 9.01, the total number of negatively charged residues (Asp+Glu) was 19, the total number of positively charged residues (Arg+Lys) was 23, and the molecular formula was C790H1274N216O226S7. GH protein of Equus przewalskiibelonged to extracellular protein, which contained growth hormone like domain and did not have relevant transmembrane structure,with a 0.35% chance of containing signal peptide. The research lays a molecular foundation for the development and utilization of GH gene in Equus przewalskii.
