随着社会人口老龄化加剧,以阿尔兹海默症(Alzheimer’s disease,AD)为代表的认知障碍疾病越来越严重地危害着人类的生命健康,带来了巨大的社会和经济负担。然而目前AD的发病机制尚未明确,也未见有效的治疗药物。此外,现有关于AD的机制研究大多数集中在高加索人群,而关于亚洲人群的研究少见报道。收集GEO公共数据库中亚洲人AD患者与正常人mRNA/miRNA芯片表达数据,筛选核心差异基因与差异miRNA,构建miRNA-mRNA差异表达网络。结果发现hsa-mi-22、hsa-miR-24、hsa-miR-125b-1-3p、hsa-mi-125-3p等差异miRNAs以及四个脑区的靶基因,并构建四个脑区的miRNA-mRNA差异表达网络。发现了四个脑区共有靶基因SLC14A1以及单脑区特有靶基因,为亚洲人AD的诊疗提供新的靶点与依据。 |

Abstract: |
With the rapid growth of the aging society, cognitive impairment diseases, represented by Alzheimers disease (AD), have been increasingly endangering human life and health, and are placing heavy burdens on society and economics. The molecular pathogenesis of AD is still not clear, and there are still no effective drug treatments for AD. While most of the existing studies on AD pathology were conducted in the Caucasian population, there were few in the Asian populations. In this study, the mRNA/miRNA expression profiles for AD patients and normal controls in the Asian population were collected from the publicly available GEO database. The miRNA-mRNA differential expression networks in four brain regions were constructed based on the differential expression analyses and target-gene predictions. Differentially expressed miRNAs, including hsa-mi-22, hsa-miR-24, hsa-miR-125b-1-3p, and hsa-mi-125-3p, and corresponding target genes in four brain regions were found. SLC14A1 was the target gene in the four brain regions, while other target genes are expressed in specific brain regions. Such target genes are expected to serve as potential new targets for the diagnosis and treatment of AD in the Asian population. |
